Abstract
Marek’s disease (MD) is a re-emerging viral disease of chickens and a serious eco-nomicthreattothepoultryindustryworldwide.Continuoussurveillancewithmolec-ularinvestigationisessentialtomonitortheemergenceofvirulentMarek’sdiseasevirus(MDV)strainsandtodeviseanyappropriatevaccinationstrategyandimplementbio-securityprogrammes.Inthepresentstudy,weinvestigatedthecasesofMDout-breaksinvaccinatedpoultryflocks.TheMDoutbreakwasconfirmedthroughnecropsy(mainlyvisceraltumours),histopathologyandviralgenespecificPCR.Thepathotypesof the field MDV strains were assessed by molecular analysis of three virulence-associatedgenes,meq,pp38andvIL-8.TheMeqsequenceofthefieldstrainsanalyzedinthisstudylackedthe59aauniquetomildstrains,indicatingthattheyarepotentiallyvirulentstrains.Mutationatposition71andthepresenceoffiveprolinerichrepeatsinthetransactivationdomain,bothassociatedwithvirulencewereobservedinthesestrains;however,thesignaturesequencesspecifictoveryvirulentplusstrainswereabsent.PhylogeneticanalysisofmeqoncogenesequencesrevealedclusteringofthefieldstrainswithNorthIndianstrainsandwithaveryvirulentplusATE2539strainfromHungary.Analysesofpp38proteinatpositions107and109andvIL-8proteinatpositions4and31showedsignaturesofvirulence.Sequenceandphylogeneticanal-ysisofoncogeneandvirulence-associatedgenesoffieldMDVsfromvaccinatedflockindicatedthatthesestrainspossessedmolecularfeaturesofvirulentstrains.
