Abstract

The affinity of aptamer is the sole reason for which they are being compared to be superior over antibodies and they have a potential to replace them as a therapeutic agent or a signaling molecule or in the field of biosensors where it has been exploited enormously nowadays. Since the advent of aptamers, although high affinity aptamers are being selected by in vitro selection, there has always been a debate over increasing the affinity of aptamers selected or to select high affinity aptamers in short time period. The various methods and techniques that are known till date for increasing affinity of an aptamer include truncation of aptamers, modification of bases, sugar ring structure, valency, etc. A short review of post-SELEX processes for increasing affinity of the aptamer and to precisely determine its affinity, various methods like surface plasmon resonance, isothermal titration calorimetry etc., are being used, has been discussed.
